MeSH: Isopropyltiogalaktosid - Finto
Supplied in one bottle containing 5 g. IPTG (Isopropyl ß-D-1-thiogalactopyranoside), is a molecular biology reagent. This compound is a molecular mimic of allolactose, a lactose metabolite that triggers transcription of the lac operon and it is therefore used to induce protein expression where the gene is under the control of the lac operator. Another less characterized element of lac operon induction is the transacetylase LacA. It is known that IPTG can be acetylated by LacA (Herzenberg, 1961) but it is not reported if this enzyme activity has an influence on lac operon induction. As a result, LacA is not considered in probably all works with lac operon elements.

IPTG is a substance which mimicks the presence of allolactose (a metabolite of lactose) and it can activate transcription from the lac operon. As IPTG (in contrast to allolactose) cannot be hydrolyzed by β-galactosidase, it's concentration in the cell stays the same. Thermo Scientific IPTG (isopropyl-beta-D-thiogalactopyranoside) is a highly stable synthetic analog of lactose. It inactivates the lac repressor and induces synthesis of beta-galactosidase, an enzyme that promotes lactose utilization. IPTG is used to induce expression of cloned genes under control of the lac operon (see Reference 1). It is used in conjunction with X-Gal (#R0941) to determine Description Gene Regulation and the Lac Operon - YouTube.

Figure 2.6.1: lac Operon lac Z codes for b-galactosidase, which is an enzyme that cleaves b-galactosides (e.g. lactose).; lac Y codes for permease, which is involved in the transport of b-galactosides into the cell.; lac A codes for b-galactoside transacetylase, which acetylates b-galactosides.; A mutation in either lac Z or lac Y can lead to a lac-genotype, i.e.

Isopropyl β-D-1-thiogalactopyranoside, abbreviated IPTG, is a molecular biology reagent. This compound is used as a molecular mimic of allolactose, a lactose metabolite that triggers transcription of the lac operon. Unlike allolactose, the sulfur (S) atom creates a chemical bond which is non-hydrolyzable by the cell, preventing the cell from “eating up” or degrading the inductant
Control of the lac operon with isopropyl β-D-1-thiogalactopyranoside (IPTG) has been used to regulate gene expression in Escherichia coli for countless applications, including metabolic engineering and recombinant protein production.

IPTG is an inducer, binding to the repressor protein and inhibit its binding to the operator region in the lac Operon and allows the lac-Operon itself to be expressed (Lab Manual, 2010). Moreover, in the tubes where the lactose was added, the amount of production …
IPTG (Isopropyl β-D-thiogalactoside, Isopropyl thiogalactoside, Isopropyl β-D-1-thiogalactopyranoside, lad-y) is a galactose and a molecular mimic of allolactose that triggers transcription of the lac operon, stimulating protein expression. 2019-1-28
2021-4-23 · We have developed a series of circuits for optogenetic regulation of the lac operon, which we call OptoLAC, to control gene expression from various IPTG-inducible promoters using only blue light.
